Zephyr AI's $111M Series A Fuels Precision Medicine Expansion

The McLean-based AI health startup raised a massive Series A in 2024, backed by Lilly and Revolution. Since then: a pharma veteran CEO, key acquisitions, and strategic partnerships.

Zephyr AI's $111M Series A Fuels Precision Medicine Expansion

The McLean, Virginia offices of Zephyr AI seemed an unlikely destination for one of last year's biggest venture checks in healthcare. But on March 13, 2024, the precision medicine startup closed $111,354,486.51 in Series A funding—a figure that placed it among the rarified tier of AI health companies commanding nine-figure rounds.

Revolution Growth led the financing. Joining the table: pharmaceutical giant Eli Lilly & Company, philanthropist Jeff Skoll, EPIQ Capital Group, and roughly 30 additional investors, according to SEC filings reviewed by the company. The round priced Series A preferred shares at approximately $17.34 apiece, issuing 6.42 million shares in total.

For a company barely two years removed from an $18.5 million seed round—led by Lerner Group Investments and M-Cor Holdings—the Series A marked something more than validation. It was a sharp turn into the spotlight for a thesis that, in 2024, still felt audacious to some observers: that machine learning models trained on vast, fragmented pools of real-world clinicogenomic data could fundamentally reshape how drugs are discovered, how patients are stratified for trials, and how diagnostics are deployed.

Zephyr planned to plow the capital into its data federation tools and ML algorithms, with an initial focus on oncology and cardiometabolic disease. The company also intended to scale both its scientific bench and commercial teams while enriching the datasets that feed its predictive engines.

Whether the bet would pay off hinged on execution—a question the company began answering 17 months later in an unexpected way.

Enter the Pharma Veteran

On August 26, 2025, Zephyr named Allen Chao its new CEO. The announcement carried weight beyond the typical executive shuffle. Chao had founded Watson Pharmaceuticals, building it into a generics powerhouse before its transformation into Actavis and eventual merger with Allergan. He'd been quietly watching Zephyr as a seed investor and board observer since 2022.

His arrival replaced interim CEO Jeff Sherman, who stepped back into his original role as chief technology officer and co-founder. Perhaps more tellingly, the leadership transition coincided with another piece of news: Zephyr had acquired Aster Insights, the company formerly known as M2Gen.

The deal brought something Zephyr badly needed—scale. Aster controlled longitudinal data from the ORIEN network (Oncology Research Information Exchange Network), part of the Total Cancer Care consortium. That meant access to lifetime-consented molecular and clinical records from more than 400,000 patients across 17 cancer centers. For a company betting on the richness of real-world evidence, it was a vault worth cracking.

Zephyr had been assembling other pieces of its leadership puzzle as well. John Applegate joined as CFO in May 2024, a few months after the Series A closed. The chief scientific officer role took longer to fill. Lisa Eli stepped into that seat in what the company described as April 2026—a timeline that suggests either forward-looking planning or the still-evolving nature of Zephyr's disclosure calendar. (The company has maintained a cadence of scientific conference presentations, including posters at the American Association for Cancer Research and the American Society of Clinical Oncology annual meetings in 2024, and has signaled continued presence at AACR in the years since.)

Partnerships As Product Strategy

Executive hires tell one story. Partnership announcements tell another—often a more revealing one.

In October 2025, Zephyr unveiled a collaboration with Guardant Health, the liquid biopsy and precision oncology diagnostics company. The arrangement pairs Guardant's multimodal molecular data with Zephyr's drug-response prediction algorithms, with integration planned into Guardant's Infinity AI platform. It's the kind of deal that suggests Zephyr sees its technology not as a standalone product but as infrastructure embedded into existing clinical and commercial workflows.

That approach traces back to earlier moves. In November 2022, shortly after emerging from stealth, Zephyr partnered with MedStar Health—which also participated in the seed round—to develop predictive analytics for chronic disease management, beginning with type 2 diabetes. Then came a March 2024 tie-up with KangarooHealth, designed to marry Zephyr's insights with remote patient monitoring capabilities.

A May 2024 profile in BioSpace mentioned NexImmune as another collaborator, though details of that relationship remain opaque.

The Products Beneath the Pitch

What, exactly, is Zephyr selling?

According to investor Red Cell Partners—which incubated the company in 2021—Zephyr offers two core products: AIM-Bx, a platform for predicting drug response, and Zephyr Nexus, which surfaces real-world clinical insights from federated data sources.

The Series A proceeds are funding work across therapeutic areas. In oncology, the company presented a composite biomarker for predicting sensitivity to PARP inhibitors at ASCO 2024. In cardiometabolic disease, the company has been quieter about specific milestones, though partnerships like the MedStar collaboration hint at ongoing development.

Zephyr lists between 11 and 50 employees on LinkedIn—a deliberately lean headcount for a company processing datasets at this scale. The bet, it seems, is that compute and partnerships can substitute for mass hiring, at least in the early innings.

Context and Questions

The $111 million round placed Zephyr among 49 U.S. AI startups that raised $100 million or more in 2024, according to a December TechCrunch analysis. That cohort included both obvious names and long shots. Which category Zephyr falls into may depend less on the elegance of its algorithms than on whether biopharma companies—and eventually payers—will adopt its tools at scale.

Precision medicine has cycled through hype curves before. The premise—match the right drug to the right patient at the right time—is clinically compelling but commercially elusive. Data fragmentation, interoperability headaches, regulatory uncertainty, and the sheer inertia of clinical practice have stalled more than one well-funded effort.

Zephyr's approach—federating disparate data sources rather than trying to centralize them, then training models that can predict outcomes without moving sensitive patient information—is designed to sidestep some of those obstacles. Whether it works remains an open question.

For now, the company has capital, partnerships, datasets, and a CEO who's built and sold a pharmaceutical company before. That's more than most startups can claim 18 months after a Series A. It's also, perhaps, just enough runway to prove whether the thesis holds—or whether the market's belief in execution was premature.
